老年人群血管内皮型一氧化氮合酶基因Glu298Asp多态性与原发性高血压病的关系 被引量:8

摘要 目的 了解中国老年人群血管内皮型一氧化氮合酶 (eNOS)基因Glu2 98Asp多态性的分布 ,探讨其与原发性高血压病的关系。方法 随机选择门诊及住院病例中确诊的原发性高血压病患者 95例为研究组 ,同时选择性别、年龄与研究组相匹配的体检正常者 95例作为对照组 ,分别测定他们的空腹血脂、身高、体重及坐位血压 ,应用聚合酶链反应 (PCR)和限制性片断长度多态性分析 (RFLP)检测eNOS基因Glu2 98Asp多态性 ,并进行两组间的比较。结果 研究组Glu Asp基因型构成比为 2 6 3 % ,对照组为 1 2 6 % ,两组eNOS基因Glu2 98Asp构成间差别有显著性意义 (χ2 =5 67,P <0 0 5)。 2 98Asp等位基因频率研究组为 1 3 2 % ,对照组为 6 3 % ,两组等位基因频率间差别亦有显著性意义 (χ2 =5 0 6 ,P <0 0 5)。结论 eNOS基因Glu2 Objective To investigate the association of Glu298Asp polymorphism of the eNOS gene with essential hypertension in old people.Methods 95 essential hypertension were randomly chosen from outpatients and inpatients as study group,and the same amount of sex,age-matched healthy people as control group.Their height,weight and blood pressure were examined and their fasting plasma lipid concentrations were measured.Glu298Asp polymorphisms of the eNOS gene were measured by using the method of PCR and RFLP.Results The constituent ratio of Genotype Glu/Asp in the study group(26 3%) was higher than that in the control group (12 6%),there was significant difference between the study group and the control group (χ 2=5 67,P<0 05).The allelic frequencies of 298Asp in the study group(13 2%) was significantly higher than that in the control group (6 3%).Comparing the allelic frequency between two groups there was significant difference (χ 2=5 06,P<0 05).Conclusion Glu298Asp variant of the eNOS gene may be an independent predictor of essential hypertension.
