摘要
目的 :探讨重组肿瘤坏死因子α(rhTNF α,TNF α)、重组粒细胞集落刺激因子 (rhG CSF ,G CSF)对脐血CD3AK细胞诱导HL 6 0细胞凋亡的影响。方法 :用细胞形态学观察 ,DNA琼脂糖电泳 ,原位末端标记法检测HL 6 0 ,CD3AK细胞诱导的HL 6 0 ,TNF α诱导的HL 6 0 ,CD3AK与TNF α共同诱导的HL 6 0 ,CD3AK与G CSF共同诱导的HL 6 0进行检查分析。结果 :HL 6 0自然凋亡率 (4.6 0± 1.71) % ,;CD3AK细胞诱导的HL 6 0凋亡率 (15 .6 0±3.2 1) % ;TNF α(5ng/ml)诱导的HL 6 0的凋亡率 (5 .2 0± 2 .0 7) % ;CD3AK与TNF α共同诱导的HL 6 0凋亡率 (36 .70± 4 .4 0 ) % (F =94 ,P <0 .0 1)。CD3AK与G CSF共同诱导的HL 6 0凋亡率 (4.88± 2 .2 3) % (F =5 8,P <0 .0 1)。结论 :TNF α协同CD3AK细胞诱导HL 6 0凋亡 ;G CSF抑制CD3AK细胞诱导HL 6 0凋亡。
Objective: To study the effect of TNF α and G CSF on the HL 60 apoptosis induced by cord blood CD3AK. Methods: Apoptosis was detected by techniques of TDT end labeling, cell morphology and DNA agarose gel electrophoresis. The percentage of apoptsis was determined in HL 60 cells, HL 60 cells induced by CD3AK cells, HL 60 induced by TNF α, HL 60 cells induced by both TNF α and CD3AK cells, as well as HL 60 induced by both G CSF and CD3AK. Results: The percentage of HL 60 cells apoptosis in the corresponding tuation mentioned above were (4.60±3.21)%, (15.60±3.21)%,(5.20±2.07)%,(36.70±4.40)%, and (4.88±2.23)% respectively (F=94,P <0.01; F=58, P<0.01). Conclusion: TNF α and cord blood CD3AK cells were synergetic in inducing HL 60 cells apoptosis. G CSF could inhibit HL 60 cells apoptosis induced by cord blood CD3AK cells.
