摘要
目的:观察中华眼镜蛇毒F组分对血小板α-颗粒膜蛋白表达的影响,确定F组分是否具有抑制血小板活化的功能。方法:实验分成对照组、凝血酶组和各给药组(凝血酶+F组分),应用酶联免疫吸附法(ELISA)测定健康成年人血浆中的α-颗粒膜蛋白(GMP-140)含量的变化。结果:凝血酶能引起血浆中的GMP-140含量明显升高,中华眼镜蛇毒F组分 100、30 μg/mL明显降低凝血酶引起的血浆GMP-140含量升高,并呈现一定程度的剂量依赖关系,两组比较P<0.05。结论:中华眼镜蛇毒F组分能够抑制激动剂引起的血小板活化。
To study the effect of fraction F of Naja Naja atra venom on expression of alpha granule membrane protein( GMP140) of platelet,and to determine whether the fraction F can inhibit the activation of platelet. Methods: three groups were included in the study: control group, thrombin group and treatment group ( thrombin + fraction F). The content of GMP140 in plasma was determined by enzyme-linked immunosorbent assay ( ELISA ). Results: Thrombin significantly increased the level of GMP140 in plasma, and the fraction F of Naja Naja atra venom remarkably inhibited the production of GMP140 induced by thrombin at dosages of 100, 30 and 10 μg/mL respectively, with a manner of dose-dependent. Significant difference was observed between two groups (P < 0. 05 ). Conclusion: Fraction F of Naja Naja atra venom can inhibit the activation of platelet induced by angonist.
