利用正交试验等设计 ,首次系统开展了 84K杨叶片外植体再生系统建立研究 ,确定了84K杨叶片外植体最适不定芽诱导培养基及激素配比为 MS+ 6- BA1 .5 mg/L+ NAA0 .1mg/L,不定芽最适生根诱导培养基及激素配比为 GMS+ NAA0 .0 1 mg/L+ IBA0 .2 5 mg/L。该实验结果为
Through the application of orthogonal designing, a systematic research into the establishment of the leaf explant regeneration systems for the poplar 84K was for the first time carried out. The optimal differentiation medium and proportion of hormone for leaf explant is MS+BA1.5 mg/L+NAA0.1 mg/L, and the optimal rooting medium and proportion of hormone for shoot is GMS+NAA0.01mg/L+IBA 0.25 mg/L. The results achieved laid a sound foundation for regeneration of poplar 84k on an industrial basis and gene transformation by means of leaf disc method.
