半球缺血沙鼠脑表达MMP-9的变化 被引量:2

摘要 目的 :动态观察沙土鼠脑缺血再灌脑内MMP - 9的蛋白表达情况 ,探讨它在脑缺血损伤中的作用。方法 :制作沙土鼠半球缺血 /再灌注模型 ,分别于术后 6 ,2 4,48h ,5d和 14d取脑 ,免疫组化技术观察MMP - 9的表达。结果 :免疫组化发现MMP - 9在正常组及手术组非缺血半球没有表达 ,缺血半球术后 6h开始表达 ,并在 2 4~ 48h表达最强 (P <0 .0 1)。结论 :脑缺血再灌诱导沙鼠脑微血管内皮细胞MMP - 9表达增加。MMP- Objective:To investigate the expression of MMP-9 after cerebral ischemia and reperfusion and their possible roles in cerebral ischemic injury.Methods:Hemispherical ischemia and reperfusion was made by clamping the common carotid artery (CCA) of Mongolian gerbils and the clip was removed after 60 minutes of occlusion.The forebrains were removed from each animal 6,24,48 hours, 5days and 14 days after surgery.MMP-9 protein expression was detected by immunohistochemistry.Results:Expression of MM-9 appeared 6 hours after ischemic hemisphere, and the strongest staining was between 24~48 hours(P<0.01).Conclusions:Increased MMP-9 expression is induced by cerebral ischemia and reperfusion of gerbil.The early increased MMP-9 may play an important role in initial cerebral ischemia-reperfusion injury.
