摘要
目的 观察大鼠脑缺血再灌注后缺血侧半球皮质和缺血核心区皮质内肿瘤坏死因子 (TNF -α)含量的变化。方法 将大鼠随机分为缺血侧半球组和缺血核心区组 ,右侧大脑中动脉闭塞 2h ,于再灌注后 0、6、16、2 4和 4 8h处死大鼠 ,用ELISA法分别测定缺血侧半球皮质和缺血核心区皮质内TNF -α含量。结果 缺血侧半球皮质内TNF -α含量于脑缺血再灌注后 6h明显升高 ,16h达高峰 ,2 4h恢复到正常水平 ,4 8h再次明显升高。缺血核心区皮质内TNF -α含量于脑缺血再灌注后无明显变化。结论 TNF -α参与了脑缺血再灌注后缺血核心区以外皮质内的损伤反应过程 ,而与缺血核心区皮质内的损伤反应可能无关。
Objective To study the concentration of TNF-α in the rat cerebral cortex in contrast between the ischemic hemisphere and the ischemic core at different times after transient focal cerebral ischemia.Methods The rats were randomly divided into the ischemic hemisphere and the ischemic core groups,and were subjected to the right middle cerebral artery occlusion for two hours,then were killed at 0,6,16,24 and 48 hours after reperfusion.The level of TNF-α was measured by enzyme linked immunosorbent assay(ELISA).Results The level of TNF-α in the cerebral cortex of the ischemic hemisphere increased significantly at six hours,peaked at 16 hours,returned to normal level at 24 hours,reached another peak at 48 hours after reperfusion,but it did not change at all after reperfusion in the cortical ischemic core.Conclusion TNF-α may contribute to the injurious reactions in the cerebral cortex of the ischemic hemisphere except the ischemic core.
