摘要
本研究自病毒介导的毒性T淋巴细胞的细胞毒水平探讨了病毒性心肌炎(VMC)的发病机制及发展为扩张型心肌病(DCM)中的作用,以及心肌细胞离子通道的改变在VMC发病中的作用;建立了VMC及DCM病毒学新检测,尤其是心肌中病毒VP_1及多病基因检测;证实肌钙蛋白I(cTnI)作为心肌损伤的较高特异性指标,自行制成试剂盒;建立了小鼠心肌炎模型用超声心动图检测心功能的方法;自小鼠心肌炎模型及全国12家医院临床VMC及DCM患者中证实用黄芪、牛磺酸、辅酶Q_(10)等综合治疗优于一般常规治疗,又自实验及DCM患者中证实加用生脉可改善心脏功能,进一步改善预后。
The role of virus mediated cytotoxic T lymphocytes was observed in the pathogenesis of viral myocarditis and the dilated cardiomyopathy succeeded, and changes in ion channel of cardiomyocytes were also investigated in the pathogenesis of viral myocarditis. Detection of enteroviral VP1 and multiple viral genes including enterovirus, HCMV、 HCV 、 etc. in myocardial tissues from patients with viral myocarditis and dilated cardiomyopathy were performed; detection of cTnI in peripheral blood as a specific marker of cardiac damage in myocarditis and cardiomyopathy was further verified and a kit for detection of cTnI was developed; a method for assessment of theart function was established using echocardiography in viral myocarditis mouse modle. Comprehensive treatment with Astragalus Membreances、 taurine and CoQ10 was experimentally and clinically proved better than routine treatment in viral myocarditis mice and patients with viral myocarditis and dilated cardiomyopathy, and routine treatment plus Sheng Mai Shan could improve heart function.
