摘要
目的 应用重组腺病毒载体介导的P2 1WAF1/CIP1基因转染血管平滑肌细胞 (VSMC) ,探讨P2 1WAF1/CIP1基因对平滑肌细胞凋亡的诱导作用。材料与方法 P2 1WAF1/CIP1的重组腺病毒载体转染体外培养的猪主动脉平滑肌细胞 ,应用RT PCR检测外源性P2 1WAF1/CIP1基因的表达 ,并通过细胞形态、原位细胞凋亡分析、流式细胞仪和细胞生长曲线等方法观察P2 1诱导VSMC凋亡 ,抑制VSMC增殖的情况。结果 P2 1WAF1/CIP1基因的重组腺病毒载体可以有效地转入平滑肌细胞中 ,诱导平滑肌细胞凋亡 ,并能显著地抑制平滑肌细胞增殖。结论 外源性P2 1WAF1/CIP1基因转移可以诱导平滑肌细胞凋亡 ,显著地抑制平滑肌细胞增殖。
Objective To investigate the induction of adenovirus mediated P21 WAF1/CIP1 in the apoptosis of vascular smooth muscle cells (VSMC). Materials and Methods A replication-defective adenovirus encoding P21 WAF1/CIP1 gene was transfected into the cultured VSMC. The expression of P21 WAF1/CIP1 gene was detected by RT-PCR methods. The apoptosis induced by P21 WAF1/CIP1 gene product was observed with light and electron microscopy, flow cytometric analysis and in situ cell apoptosis detection.Results The P21 WAF1/CIP1 gene could be effectively transferred into VSMC by adenovirus vector and its product could induce apoptosis of transfected VSMC.Conclusion The P21 WAF1/CIP1 gene transfection can induce the apoptosis of VSMC and markedly inhibit the multiplication of VSMC.
