摘要
目的 探讨奥曲肽是否通过诱导人胆管癌细胞凋亡而抑制肿瘤生长 ,以及与细胞内游离钙的关系。方法 采用人胆管癌细胞系SK ChA 1,应用生长曲线 ,片段DNA琼脂糖凝胶电泳 ,AnnexinV FITC技术及激光共聚焦显微镜技术等方法进行检测和观察。结果 在奥曲肽作用下SK ChA 1细胞生长受抑制。奥曲肽处理SK ChA 1细胞后 ,AnnexinV标记的凋亡细胞增多 ,DNA凝胶电泳显示典型的凋亡梯状图谱。细胞凋亡的同时细胞内游离钙浓度升高。结论 奥曲肽通过细胞内游离钙的变化 ,诱导人胆管癌细胞凋亡 ,是奥曲肽抑制胆管癌生长的机理之一。
Objective To investigate whether apoptosis induced by octreotide resulted in human cholangiocarcinoma growth inhibition and the relation between apoptosis and intracellular free calcium in vitro.Methods Proliferation curve, agarose gel electrophoresis.Annexin V FITC and laser scanning confocal microscopy technique were performed to identify the inhibitory effect on cell proliferation and the induction of apoptosis in human cholangiocarcinoma cells(CK ChA 1).Results Octreotide significantly reduced the SK ChA 1 cell growth by serum in long experiments. The cells treated with octreotide increased annexin V positive cells.DNA of treated SK ChA 1 cells appeared a ladder pattern characteristic of apoptosis.Meanwhile the intracellular free calcium concentration was increased.Conclusion Octreotide could inhibit the proliferation activity and induce apoptosis of cholangiocarcinoma cell line SK ChA 1.The results suggest that apoptosis followed by change of intracellular free calcium concentration is one of the mechanism of octreotide in inhibition of cholangiocarcinoma growth.
