摘要
目的 :探讨连云港地区原发性胃腺癌 P5 3基因变异发生情况。方法 :应用聚合酶链反应单链构象多态分析法(PCR- SSCP)对 2 0例原发性胃腺癌的 P5 3基因第 7、8外显子突变进行了检测。结果 :共发现突变 5例 (2 5 % ) ,其中第 7外显子突变 4例 ,第 8外显子突变 1例。在 2 0例原发性胃腺癌组织分型中 ,高分化和中分化各 3例 ,均未见 P5 3基因第 7、8外显子突变 ;低分化 13例 ,发现 P5 3基因第 7外显子突变 3例 ,第 8外显子突变 1例 ;低分化 -未分化 1例 ,亦发现第 7外显子突变。结论 :P5 3基因突变与原发性胃腺癌发生有关 。
Objective:To study the 7th and the 8th exon mutation of P53 gene in primary gastric adenocarcinoma. Methods:PCR-SSCP was taken to examine the 7th and 8th exon mutation of P53 gene in 20 cases of primary gastric adenocarcinoma.Results:5 cases of the 7th and 8th exon mutation of P53 gene were found(25%),3 cases of the 7th exon mutation and one case of the 8th exon mutation.Among 20 cases of primary gastric adenocarcinoma,there were 3 cases of well-differentiated ones and 3 cases of middle-differentiated ones in which the 7th and 8th exon mutation of P53 gene were not found;13 cases of poorly-differentiated ones in which 3 cases of the 7th exon mutation and one case of the 8th exon mutation of P53 gene were found;one case of poorly-undifferentiated ptimary gastric andenocarcinoma which showed the 7th exon mutation of P53 gene.Conclusion:The mutation of P53 gene is correlated to primary gastric andenocarcinoma,which is of instructive importance to the treatment of primary gastric andenocarcinoma.
