摘要
目的 :建立迟发性超敏反应的新西兰兔动物模型并研究超敏反应对假体周围组织及滑膜炎症反应的影响。方法 :通过皮内注射K2 Cr2 O7和福氏完全佐剂以及涂皮相结合的方法在新西兰兔体内建立对Cr6+ 产生迟发性过敏反应的动物模型 ,在右侧股骨髁内植入钛合金棒 ,并通过关节腔间断注射Cr6+ ,在术后 8周时检测致敏情况并观察膝关节滑膜和假体周围的炎症反应。结果 :有 4/6只兔对Cr6+ 产生了迟发性过敏反应 ,在致敏组中 ,滑膜和假体周围骨髓中炎症细胞浸润明显增加 ,局部形成典型的肉芽肿性组织 ,并可见骨髓中破骨细胞数量的增加。结论 :在致敏状态下 ,植入假体后可使假体周围炎症反应明显增强 。
Objective:To establish the Cr 6+ sensitivity animal models and investigate the effects of hypersensitivity on tissue reaction around prosthesis. Methods:Eight rabbits were immunized by repeated injection of potassium dichromate (K 2Cr 2O 7)following an initial injection of chromium in Freund's complete adjuvants(FCA).Then Ti alloy plugs were implanted in the femur of rabbits,and Cr 6+ were injected into right knee joints repeatedly every two weeks.After eight weeks,the lymphocyte transfer test (LTT) was done to examine the hypersensitivity and the histology of synovialis and tissue around plugs were evaluated.Results:There were four rabbits being sensitized by Cr 6+ .The tissue reaction to plugs and synovialis in sensitized animals show evident granoma with a significant increase in inflammatory cells and foreign body giant cells when compared with the reaction in non sensitive animals.Also,we found an increase of osteoclasts around prostheses in the sensitized animals.Conclusion:The hypersensitivity can increase the inflammation reaction of synovial membrane and tissue around implanted plugs,also lead to bone resorption.
