摘要
目的:通过抗凋亡基因bcl-xL在正常肝和肝癌组织中的表达及与肝癌患者临床资料关系分析,从分子水平探讨该基因控制肝癌细胞凋亡的作用。 方法:应用Northern blot分析法对正常肝和肝癌组织中bcl-xLmRNA表达水平进行测定,并对肝癌患者bcl-xL mRNA表达水平同临床资料关系行统计学分析。 结果:Northern blot分析显示,与正常肝组织相比,95%(20/21)肝癌组织bcl-xL mRNA明显呈低表达,密度测定分析法显示:肝癌组织bcl-xL mRNA水平(2.43±2.22)较正常肝组织(8.50±5.74)低3.5倍(P=0.0002)。肝癌组织中bcl-xL mRNA水平同患者年龄、性别、肿瘤分化程度和分期无关。 结论:抗凋亡基因bcLxL在正常肝和肝癌组织中的不同表达提示该基因在此癌细胞凋亡中发挥重要作用,且与肝癌发展的不同阶段尚无明显相关。
AIM:To investigate the effect of bcl-xL on the apoptosis of hepatocellular carcinoma, by means of analyzing the expression of this gene in the tissues of normal liver and hepatocellular carcinoma and its relationship with clinical parameters in patients with the carcinoma.METHODS: Expression of bcl-xL mRNA in normal liver and hepatocellular carcinoma was measured by Northern blot. The relationship between the expression of bcl-xL mRNA and clinical parameters of patients with the carcinoma was statistically analyzed.RESULTS: Northern blot analysis indicated that 95 %(20/21) of hepatocellular carcinoma tissues expressed bcl-xL at low level and 38 % of them exhibited faint expression of bcl-xL, in contrast with the normal liver tissues. Densitometric analysis revealed that hepatocellular carcinoma exhibited a mean 3.5-fold decrease (2.43 ±2.22) in bcl-xL mRNA levels in comparison with normal controls (8.50 ±5.74) (P = 0. 0002). There was no correlation between the expression of bcl-xL mRNA and age and gender of the patients, tumor differentiation and stage.CONCLUSION: This finding is consistent with the fact that apoptosis rarely occurs in normal liver but increases in hepatocellular carcinoma and indicates that bcl-xL may play a very important role in regulating the apoptosis of normal liver and the carcinoma. But bcl-xL does not influence the differentiation and invasiveness of the tumor.
