The Impact of Endothelin-1 on Spinal Cord Blood Flow and Blood-Spinal Cord Barrier in Normal Spinal Cord in Rats

摘要 目的观察外源性内皮素 -1(ET-1)对正常脊髓血流及血脊屏障的影响。方法大鼠蛛网膜下腔注射10ngET-1或生理盐水。激光多普勒血流仪监测脊髓血流 ,伊文思兰 (EB)评价血脊屏障功能。干湿重法测定脊髓中水含量。结果ET-110ng鞘内注射明显减少脊髓血流至少达4h,注射ET-115min后各时点脊髓血流与对照组比较差异有显著性意义(P<0.05),注射ET-1组脊髓中EB与水含量高于对照组 ,两者比较差异有显著性意义(P<0.01)。结论鞘内注射ET-1能引起正常大鼠脊髓缺血 ,破坏血脊屏障。 Objective To observe the impact of exogenous endothelin_1(ET_1) on spinal cord blood blow(SCBF) and blood_spinal cord barrier(BSCB) in normal spinal cord in rats.Methods Saline or ET_1(10ng) was administered into subarachnoid space. A laser dopplor flowmetry was utilized for measuring SCBF. The permeability of BSCB was evaluated quantitatively using Evans blue. Tissue edema was determined by percentage water content.Results Intrathecal administration of 10ng ET_1 had SCBF decrease for at least 4 hours. There was a significant decrease in SCBF compared with control 15 minutes after administration(P<0.05).ET_1 increased the contents of Evans blue and water in the cord of normal rat. There was a significant increase in content of Evans blue and water compared with control(P<0.01). Conclusions Intrathecal administration of ET_1 can cause ischemia of the cord and breakdown of BSCB in normal rats .ET may play a role in ischemia and breach of BSCB following SCI.
