微囊藻毒素LR在动物体内整体水平及细胞水平的分布 被引量:18

摘要 目的 研究和探讨微囊藻毒素LR(MicrocystinsLR ,MCLR)在机体内分布的靶器官及其在靶器官中的细胞水平定位。方法 用1 2 5I MCLR标记物纯化 ,并注入小鼠体内 ,分别经同位素示踪技术和放射性自显影方法研究1 2 5I MCLR在小鼠体内整体水平和细胞水平的分布。结果 同位素示踪显示 ,静脉注射、腹腔注射和口服 3种不同途径进入体内的1 2 5I MCLR ,70 %以上分布在肝脏和肾脏 ;提示这两个脏器是它的靶器官。放射性自显影研究表明 ,1 2 5I MCLR在靶器官肝脏和肾脏内分别定位于肝细胞核内和肾皮质的肾细胞核内。结论 该研究结果将为进一步探讨MCLR的损伤机制提供理论依据。 Objective The distribution of Microcystin LR (MCLR)in mice and the further cellular locations in the target organs were studied. Methods First,MCLR was labeled with radioisotope 125 iodine ( 125 I),then, 125 I-MCLR was purified and given to mice via oral,intraperitoneal and vena administration.The target tissues of MCLR distribution and the cellular location of MCLR were studied via radioiso ope trace and autoradiography techniques .Results The radioisotope trace results showed that 70%radiolabeled MCLR distributed in the liver and kidney which might be the two target tissues for MCLR in mice .Autoradigraphy results indicated that MCLR was disrtibuted in the liver cell nuclei and the renal cell nuclei.Conclusion Above results may provide the theory bases to further study the damage mechanism of MCLR.
