甘氨酸在肝硬化发生发展过程中对肝组织CD_(14)基因及蛋白表达的影响 被引量:5

摘要 目的 观察甘氨酸在肝硬化形成过程中对肝组织表达CD_(14)基因和CD_(14)蛋白的影响。方法 采用复合因素建立肝硬化动物模型的同时,实验组分别用甘氨酸液(1g/d)给大鼠灌胃,或用含5%甘氨酸食物喂养大鼠,并于实验的2周未、4周末和8周末以逆转录多聚酶链反应(RT-PCR)和 western blot法检测大鼠肝组织 CD_(l4)mRNA和CD_(14)蛋白的表达。结果 在用含甘氨酸食物喂养的脂肪肝和肝硬化大鼠中,其肝组织CD_(14) mRNA及CD_(14)蛋白的表达较对照组大鼠明显减弱,其中用含甘酸食物喂养8周未的肝硬化大鼠肝组织的CD_(14) mRNA及CD_(14)蛋白表达最弱。结论 甘氨酸可下调肝组织CD_(14)基因和CD_(14)蛋白的表达。 ve To observe the effect of glycine on the expression of CD_(14) mRNA and protein of hepatictissue in the course of developing cirrhosis of rats. Methods The cirrhotic model of Wistar rats was established by com-plex pathogens. who were respectively fed with control diets and control diets adding glycine (lg/d. giving by intragastricinfusion) or 5% glycine containing diets at the same time. The rats were sacrificed at 2, 4, and 8 weeks, respectively. Hepatictissues were collected to measure the expression of CD_(l4) mRNA and CD_(l4) protein by the reverse transcriptase - polymerasechain reaction (RT-PCR) and western blot analysis. Results The expression of CD_(l4) mRNA and CD_(l4) protein in the he-patic tissue of fatty liver and cirrhotic rats fed with diets containing glycine was weaker than their control groups, and theexpression of CD_(l4) mRNA and protein was the weakest in 8 weeks cirrhotic rats fed with the diets. Conclusions G1ycinecan markedly downregulate the expression of CD_(l4) mRNA and CD_(14) protein in hepatic tissues of cirrhotic rats.
