摘要
目的 :探讨前列腺素E2 (PGE2 )免疫干预对急性坏死性胰腺炎 (ANP)大鼠肿瘤坏死因子 (TNF α)和白介素 10 (IL 10 )与肺组织超微结构改变的影响。方法 :SD大鼠 91只分成 4组 :手术对照组 (n =7)、ANP组、PGE2 前干预组和PGE2 后干预组 (每组n =2 8) ,后 3组又随机分 1h、3h、6h、12h时段 ,每时段 7只。大鼠ANP模型用 5 %牛磺胆酸钠诱导制作 ,PGE2 前干预组和后干预组分别在ANP模型制作前 2h和模型完成时肌肉注射 15 0 μg/kg和 15 0 μg/kgPGE2 。采用ELISA检测各时段大鼠血清的TNF α与IL 10含量 ,观察PGE2 前干预、后干预对大鼠血清TNF α与IL 10含量动态变化 ,同时随机取NAP组和PGE2 干预组 6h和 12h时段肺组织 ,电镜下观察超微结构改变。结果 :PGE2 前、后干预组IL 10含量峰值分别在 3h和 6h时段 ,比ANP组显著升高 (P <0 .0 0 1) ,所有时段 (1h、3h、6h、12h)TNF α均比ANP组大鼠显著降低 (P <0 .0 0 1) ,IL 10上升幅度随PGE2 给予的早晚而不同 ,TNF α亦随IL 10上升而下降。PGE2 免疫干预组与ANP组相比 ,6h时段的肺组织病变有所减轻 ,12h时段明显改善。结论 :PGE2 在上调大鼠血清IL 10、降低TNF α含量的同时 。
Objective:To study of immuno-intervention with prostaglandin E 2(PGE 2) on interleukin-10(IL-10),tumor necrosis factor(TNF-α)and pathological changes of lung in rats with acute necrotizing pancreatitis(ANP).Methods:Ninety one SD rats were divided into 4 groups:operation control group(n=7),ANP group,pre-intervention group,and post-intervention group(n=28,respectively).They were randomized redivide into different time points behind three groups (1h, 3h,6h,12h,n=7,respectively).ANP model of rat was induced by retrograde injection of 5% sodium taurocholate in the pancreatic duct.Pre-intervention group by intramuscularly injected 150 μg/kg PGE 2 before 2 hours induction of ANP,post-intervention group of PGE 2 administration were the same above dose after ANP model immediately the serum levels of IL-10 and TNF-α in rats were measured by specific enzyme linked immuno-asorbent assay(ELISA) at different time points,and some of these lung tissues were made ultrathin section and examined under electron microscopy (6h and 12h),respectively.Results:The serum IL-10 level increases in either pre-intervention group (3h) with PGE 2or post-intervention group (6h)with PGE 2,compared with ANP group were significant difference (P<0.001,respectively).Serum TNF-α level in group treated with PGE 2 compared with control group (ANP) were significant reduction(P<0.001).This result correlates well with ultrastructure findings of pulmonary injury.Compared with pulmonary constitution lesion degrees both groups of immuno-intervention with PGE 2 and control relieve at 6h and significant amelioration in 12 hours.Conclusion:This study shows that PGE 2 not only increased IL-10 concentration in the serum but also potent inhibit from TNF-α, and that PGE 2 has important regulatory effects on immunological and inflammatory responses to cause lung injury improved significantly after treatment with PGE 2.
