微囊化人胎胰岛功能的体外观察

摘要 目的 :观察微囊的通透性及对微囊化胰岛分泌功能的影响。方法 :将用胶原酶消化获得的胰岛细胞分为 2份 ,1份微囊包膜 ,1份未予包膜作为对照 ,培养 7天 ,比较两组培养液中胰岛素含量并分别用2 .7mmol·L-1、16 .6mmol·L-1浓度的葡萄糖及 16 .6mmol·L-1的葡萄糖加 10mmol·L-1茶碱刺激胰岛B细胞 ,观察其胰岛素分泌功能。结果 :两组在培养液中胰岛素含量及葡萄糖、茶碱刺激的胰岛B细胞胰岛素分泌方面无显著性差异 (P >0 .0 5 )。结论 :该微囊具有良好的通透性 。 Objective:To observe the permeability of the microcapsules and the secretic function of the microencapsulated human fetal pancreatic islets.Methods:Human fetal pancreatic islets were isolated from the embryos which were obtained from legal abortion (gestational ages 16~24 weeks) with collagenase and were divided into 2 groups.Group 1 was microencapsulated,and group 2 was free.Both groups of islets were cultured for one week,and samples were taken for the measurement of insulin concentritions on the 1st,3rd,5th or 7th day.Results:The insulin concentritions of group 1 were 1331.6±53.35,2833.43±34.61,1821.97±54.15,1320.9±31.57 μIU·ml -1 versus group 2 1282.3±22.52,2842.43±28.96,1736.4±37.26,1246.4±42.68μIU·ml -1.No significant differences were found (P>0.05)between the two groups.The insulin concentritions stimulated by glucose(concentration 2.7mmol/L,16.6mmol/L)and glucose +isobutylmethylxanthine(glucose 16.6mmol.L -1+isobutylmethylxanthine 10mmol·L -1)were 83.1±5.77,154.47±7.88,232.03±9.36μIU·ml -1 in group 1 versus 81.43±2.08,146.6±7.47,234.47±9.06μIU·ml -1 in group 2 and also no significant differences were found (P>0.05).Conclusion:These results indicate that both the permeability and the secretic function of the microencapsulated islets are not affected by the processing.
