摘要
探讨加味温胆汤对日本快速老化模型鼠(SAM-P/10)肝细胞核活性基因的影响。方法:采用DNA酶Ⅰ为活性基因探针,酶解DNA。结果:经治疗后各组肝细胞核基因组经DNA酶消化的肝细胞核酶解片段缩小,接近R/1系水平。结论:加味温胆汤具有提高SAM-P/10鼠肝细胞核活性基因水平,并增加转录活性的作用。推测其机制与其改善了SAM-P/10鼠肝细胞染色质构象,从而有利于DNA酶的消化作用有关。
To explore the influence of Jiawei Wendan prescription on the active gene in nucleus of the liver. Methods: To use active gene probe DNase I to enzomolysis DNA. Results: Active gene in nucleus of the liver are digested by DNase Ⅰ, the enzomolysis section of the treating groups in nucleus of the liver reduced to the level of SAM-R/ 1, especially nucleus. Conclusion: It shows that prescription can inrcease the level of active gene in nucleus of the liver, and increase the copying activities. The mechanism is possible that prescription improve the structure of chromatin. which is benefit to digestion of DNase Ⅰ.
