摘要
2 0 0 0年 8月于辽宁省大连长海县自然海区采得 3月龄栉孔扇贝稚贝 ,利用聚丙烯酰胺凝胶电泳技术对其进行了几种同工酶的生化遗传分析 ,得到了基本稳定的酶谱 ,同时与成贝进行了比较讨论 .共记录了 10个酶位点 2 0个等位基因 .3月龄稚贝已经存在较完善的酶系统 ,能够完成基本的新陈代谢活动 .与成贝相比 ,稚贝各酶的位点数和等位基因数都较少 ,多样性也较低 .这是与其发育阶段。
About 200 scallops ( Chlamys farreri) of 3 month age were collected from the natural seawaters of Dalian, China in Aug., 2000.The samples were lively dissected , and stored at -70℃ for use.Ten isozymes (MDH?ADH?G 6 PDH?IDH?GDH?ME?ACP?SOD?SDH?EST) were analyzed by vertical polyacrylamide gel electrophoresis (PAGE). Treated by homogenizing in icebath and centrifuging at 4℃,13 000 rad/min, the supernatant were stored at -20℃ for electrophoresis (buffer systerm:Tris Gly,pH8.3, concentrated gel(T=3.6%,pH6.7), separated gel(T=8.2%,pH8.9)).The biochemical genetic traits of 10 isozymes, including their locus number, structure, alleles, etc., were determined. 10 gene loci and 20 alleles were recorded, among which the expression of 6 loci including Est 1, Me 2, Sod 1, ACP,ADH, SDH was singlet, the rest were polymorphic.
