摘要
采用MTT法、光镜、电镜、流式细胞仪和免疫组化方法研究了^(188)Re诱导三种肿瘤细胞凋亡的形态学变化、剂量时间效应关系以及凋亡相关基因Bcl-2和Bax在其中的表达情况。结果表明:^(188)Re对三种肿瘤细胞有明显的杀伤效应,能诱导其产生凋亡的形态学变化;并且随着浓度的增大和时间的延长,凋亡率增加,Bcl-2表达减弱,Bax表达增强,二者比值下降;细胞阻滞于G_2/M期;三种肿瘤细胞对^(188)Re的敏感性由高到低依次为PC-3细胞、ER-75-30细胞、A549细胞。
Three different tumor cell lines' apoptosis induced by 188Re radiation is detected by MTT assay, light microscope, transmissional, electron microscope and flow cytometer, and the expression of Bcl-2 protein and Bax protein during the apoptosis are detected by image method. The results indicate that the three kinds of tumor cell lines can be killed by 188Re radiation with dose-effect relationship. With rise of radiation dose and prolongation of culture time, the tumor cell apoptosis rate is increased, the expression of Bcl-2 is decreased, and that of Bax enhanced, which causing the ratio is reduced. The cells are arrested in G2/M phase; the three kinds of cell are very different in sensitivity to 188Re radiation from PC-3 cell higher to A549 cell lower.
