基质金属蛋白酶-9及其抑制剂在哮喘豚鼠支气管肺泡灌洗液中的表达

摘要 目的 :观察实验性哮喘豚鼠与对照组豚鼠支气管肺泡灌洗液 (bronchoalveolarlavagefluid ,BALF)中MMP 9、TIMP 1的表达。方法 :用卵蛋白制备哮喘模型 ,免疫组织化学SABC法染色 ,苏木素复染。显微镜下观察哮喘组及对照组BALF中细胞总分数 ;MMP 9、TIMP 1阳性细胞百分比。结果 :BALF中哮喘组嗜酸性粒细胞明显高于对照组 (P <0 .0 5 ) ;巨噬细胞、淋巴细胞、嗜酸性粒细胞、中性粒细胞MMP 9、TIMP 1均呈阳性。且哮喘组MMP 9及TIMP 1阳性细胞百分比比对照组MMP 9及TIMP 1明显增高 (P <0 .0 0 1 )。结论 :MMP 9、TIMP 1可能与哮喘炎症有关。 Objective: To observe the expression of matrix metalloproteinases (MMP-9) and its inhibitor of matrix metalloproteinases(TIMP-1) in bronchoalveolar lavage fluid (BALF) of experimental and control groups. Methods: Asthmatic models were made by egg albumin. Immunohistochemical staining and counter staining with campeachy were used. Cell counts in BALF and positive cell percent with MMP-9, TIMP-1 were observed under microscope. Results: Eosinophilic granulocytes in BALF of asthmatic group is higher obviously than those in control group (P<0.05). MMP-9 and TIMP-1 in macrophagocyte, lymphocyte, eosinophilic granulocyte and netrophilic granulocyte were positive. And positive cell percent with MMP-9 and TIMP-1 in asthmatic group is higher obviously than those in control group(P<0.001). Conclusion: MMP-9 and TIMP-1 is relative to the inflammation of asthma.
