摘要
用 RAPD- PCR技术 ,对采自新疆各地的 2 7个棉花黄萎菌株的遗传分化及致病性进行了研究。从 1 2 0个随机引物中筛选出 1 1个随机引物 ,共在 2 0 4个位点上扩增出 RAPD谱带 ,其中 1 88个为多态性位点 ,占 92 .1 6%。RAPD电泳条带差异表明 ,新疆棉花黄萎病菌种群内部存在着显著的遗传分化。对 RAPD谱带进行聚类分析 ,将 2 7个菌株划分成 3大类群 ,这 3大类群与根据鉴别寄主反应划分的不同致病类型有一定的相关性 。
Genetic differentiation and pathogenicity of 27 cotton Verticillium dahliae isolates collected in Xinjiang were studied with RAPD PCR technique.11 random primers were selected from 120 ones.RAPD bands were amplified at 204 sites,188 sites were polymorphic (92.16%). Difference of RAPD banding patterns showed that there was significant genetic diversity within and between population of cotton Verticillium dahliae isolates in Xinjiang.By the cluster analysis, 27 cotton Verticillium dahliae isolates could be classified into three groups, which showed some correlations with different pathotypes classified by response of identification host. Difference of banding patterns had some relations with the locality of isolates.
