摘要
目的 探讨荷瘤大鼠 6 0 Coγ射线局部照射时血液学氧化损伤以及复合抗氧化剂抗损伤作用 .方法 采用 Walk-er- 2 5 6肿瘤细胞株接种大鼠皮下 ,得到实体瘤 ,摘除实体瘤分割成小块植入大鼠的右后腿皮下 ,制成大鼠荷瘤模型 ,将荷瘤大鼠分成 3组 ,分别为肿瘤组、照射组和抗氧化剂保护组 ,同时选取正常大鼠作为阴性对照组 .抗氧化剂保护组每日给予复合抗氧化剂 ig,分次对照射组和抗氧化剂保护组的荷瘤大鼠进行 6 0 Coγ射线局部照射 4次 ,每次间隔 1wk,累计剂量为4 7Gy,最后一次照射后 7d,活杀大鼠 ,取全血和血清测定过氧化氢酶 (CAT )、丙二醛 (MDA)、总超氧化物歧化酶 (T-SOD)和总蛋白 .结果 照射组的 MDA含量显著高于非照射组和抗氧化剂保护组 (347μmol· g- 1 vs 2 92 ,30 5和 196μmol· g- 1 ,P<0 .0 5 ) ,复合抗氧化剂显示出良好的保护作用 .而照射组的抗氧化酶类如总超氧化物歧化酶 (T- SOD)活性显著低于非照射组和抗氧化剂保护组 (3.6 5μkat· g- 1 vs4 .0 5 ,4 .82和 4 .70 μkat· g- 1 ,P<0 .0 5 ) ,CAT的活性在照射组显著低于肿瘤组和抗氧化剂保护组 (36 2 K· g- 1 vs 6 77和 6 72 K· g- 1 ,P<0 .0 5 ) .结论 6 0 Coγ射线局部照射可以产生自由基引发的全身氧化损伤 ,高效的复合抗氧化?
AIM To observe the oxidative injury of rats with tumor by local 60 Co γ irradiation and the protection of antioxidants compound. METHODS The SD rats were divided into control group as negative control group (NCG) and experimental groups (TG, RG and PG). The tumor cell strain Walker 256 was injected into the rat to get the entity tumors which were cut into small pieces and were embedded into the right rear buttocks under skin of rats. As the embedded tumor had grown up successfully, they were randomly divided into 3 groups which were tumor group (TG), radiation group (RG) and radiation plus protection group (PG). The PG was given antioxidants compound by gavage and other groups were given the same dosage water. RG and PG were locally γ irradiated with the total doses of 47 Gy, then all the rats were sacrificed and the bloods collected to measure the content of serum malondialdehyde (MDA), the activities of the total superoxide dismutase (T SOD) and the catalase (CAT). RESULTS The content of MDA in RG is much higher than that in all the other groups (347 μmol·g -1 vs 292, 305 and 196 μmol·g -1 , P <0.05). The activities of SOD and CAT in RG are much lower than those in the TG and RPG (3.65 μkat·g -1 vs 4.05, 4.82 and 4.70 μkat·g -1 , 362 K·g -1 vs 677 and 672 K·g -1 , P <0.05). CONCLUSION Local radiation can cause the oxidative injury on the domestic system of the rats and the antioxidant compound shows a protective effect against this injury.
