海鞘G蛋白偶联受体激酶cDNA的克隆

摘要 目的 从海鞘体内分离可能存在的编码G蛋白偶联受体激酶 (GRK)的cDNA克隆 ,确定其分子特征、研究其转录产物表达情况。方法 采用逆转录 聚合酶链反应 (RT PCR)、序列分析、3′RACE等方法。结果 得到了海鞘的一种GRKcDNA克隆 ,将其命名为Ci GRK1。从序列分析及分子遗传分析的结果可以看出Ci GRK1与人类GRK5的同源性最高 ,其次是果蝇GRK2。Ci GRK1mRNA在海鞘胚胎发育的整个过程中及其在幼虫体内均有表达。结论 Ci Objective To isolated and characterized cDNA clones encoding GRK from the ascidian Ciona intestinalis. Methods RT-PCR, 3′ RACE and DNA sequencing were used to identify and cloning of the GRK cDNA. Results A GRK-like gene, designated Ci-GRK1, was found in a primitive chordate, the ascidian Ciona intestinalis. The amino acid sequence of this Ci-GRK1 showed the highest homology with the human GRK5. It was also found that Ci-GRK1 was expressed at all stages of embryo development and in the larvae stage. Conclusion These results suggested that Ci-GRK1 might play an important role for a variety of developmental and physiological events of ascidian.
