光合细菌产生5-氨基乙酰丙酸(ALA)的研究 被引量:24

摘要 从 36个光合细菌菌株中筛选出的 7株紫色非硫红假单胞菌有产 ALA能力 ,其中菌株 99- 2 8ALA产量最高 .用紫外线对菌株 99- 2 8进行诱变处理 ,筛选出产量比野生菌株高 1倍多的菌株 L- 1.对影响菌株的生长和 AL A产量的因子进行了探索 .抑制剂加入时间和加入次数对 AL A产量有显著影响 .在最佳条件下 (p H7.5 ,培养对数生长期加入 AL A脱水酶抑制剂乙酰丙酸 30 mmol,AL A生物合成前体甘氨酸 30 mmol和琥珀酸 30 mmol,30 0 0 lx光照 ) ,菌株 L - 1的 AL A产量可达 2 2 .15 mg/ L. In the laboratory 7 strains of rhodopseudomonas sp were selected from 36 photosynthetic bacteria strains. Rhodopseudomonas sp strain 99-28 had the highest ALA production in these 7 strains. Rhodopseudomonas sp 99-28 strain was mutated using ultraviolet radiation and a mutant strain L-1 was obtained, in which ALA production was higher than wild strain 99-28 by about 100%. The elements which affect strain 99-28 and L-1 ALA formation were studied. Under the optimal condition of ALA formation (pH of 7.5, in the condition of supplementation of ALAD inhibitor LA and precursors glycine and succinat, and light 3 000 lx) ALA formation of mutant L-1 was as high as 22.15 mg/L.
