摘要
目的 :建立 BAL B/ c小鼠实验性自身免疫性心肌炎模型。 方法 :采用差速离心、分级饱和硫酸铵沉淀和 DEAE-Sephadex A - 5 0离子交换层析纯化等方法自猪心中提取并纯化心肌肌凝蛋白 ,经 SDS- PAGE和 Western印迹方法鉴定。以此蛋白免疫遗传易感的 BAL B/ c小鼠 ,建立自身免疫性心肌炎模型。 结果 :97% (35 / 36 )小鼠在免疫后第 14天出现不同程度的心肌炎病理改变 ,主要表现为心肌间质单个核细胞浸润、肌纤维排列紊乱和肌浆溶解。免疫鼠血清肌钙蛋白 I水平升高 ,并出现较高滴度的抗肌凝蛋白自身抗体。 结论 :猪心肌肌凝蛋白可以诱导 BAL B/ c小鼠出现典型的自身免疫性心肌炎。
Objective:To establish animal model of experimentally autoimmune myocarditis in BALB/c mice. Methods:Cardiac myosin was isolated from pig heart through methods of differential centrifugation and (NH 4) 2SO 4 fractionation, and was purified by DEAE Sephadex A 50 chromatography, judged by SDS PAGE assay and Western blotting.Then it was used for immunization of genetically predisposed mice to establish animal model of autoimmune myocarditis. Results:On the 14th day after the first immunization with cardiac myosin 97%(35/36), the BALB/c mice developed histological myocarditis with pathological features of infiltration of mononucleocytes,derangement of muscle fibers and even sarcoplasmic lysis.A high titer of myosin specific autoantibodies(IgG) and elevated troponin I level were detected in immunized mice. Conclusion:Cardiac myosin isolated from pig is capable of inducing autoimmune myocarditis in BALB/c mice.
