摘要 目的 通过胚鼠皮质神经元的体外培养 ,观察遭受缺氧刺激时 ,脑源性神经营养因子 (BDNF)对神经元的保护作用。方法 用台盼蓝染色法动态观察培养神经元的存活率及加入BDNF对生理培养状态的皮质神经元的作用 ;同时用四氮唑盐 (MTT)比色法观察不同缺氧时间实验组及对照组神经元活性的差异。结果 BDNF对正常皮质神经元有维持存活效应 ;对缺氧神经元有确切保护作用 ,该保护作用有时间依赖性 ,缺氧前2 4h加入BDNF对缺氧神经元的保护效果较缺氧即刻加入为好。结论 体外实验证实BDNF可维持皮质神经元存活 。 Objectives To observe the role of brain derived neurotrophic factor (BDNF) in hypoxia induced cortical neurons to provide new laboratory evidence for further application of BDNF in clinical practice. Methods The survival rates of cultural fetal cortical neurons were tested and the influence of adding BDNF to the culture medium on survival rates was observed though trypanblue dye; the activity of the cortical neurons was detected using MTT. Results We observed that BDNF could protect the cortical neurons from hypoxia induced neurotoxicity by preincubating the neurons before 24 hours with BDNF and adding BDNF to the cultured neurons just before exposing them to hypoxic cultural environment.The survival rate of the cortical neurons was significantly reduced after exposing to hypoxia environment, while pre incubation of the cortical neurons with BDNF before 24hr and just before exposing to hypoxia showed much higher survival rates than that of the control group in terms of preventing hypoxia neurotoxicity. So this protection was time dependent.Conclusions In vitro, BDNF plays an important role in preventing cortical neurons from hypoxia induced neurotoxicity.
