摘要
目的 :探讨碱性成纤维细胞生长因子 (bFGF)对体外培养的人牙周膜细胞 (PDLC)增殖和细胞周期的影响。方法 :采用细胞培养、MTT比色测定法和流式细胞仪 (FCM )分析技术。结果 :bFGF能明显促进人PDLC的增殖 ,且呈剂量依赖性 ;FCM对细胞周期时相分析结果显示 ,经bFGF作用后 ,人PDLC的DNA合成量 (S % )显著升高 ,G1%降低 ,反映细胞增殖活力的增殖指数PrI值 (S +G2 M ) %增高。结论 :bFGF能促进人PDLC的增殖和DNA合成 。
AIM: To evaluate the effects of basic fibroblast growth factor(bFGF) on the proliferation and cell cycle of human periodontal ligament cells(PDLC). METHODS: Cell culture technique, MTT colorimetric assay and flow cytometry(FCM) were used. RESULTS: bFGF enhanced the proliferative responses of PDLC in a dose-dependent manner. Employing the FCM technique, we observed the increased S%, the decreased G 1 % and the enhanced (S+G 2M)% when the cells were stimulated by bFGF. CONCLUSION: bFGF enhances the proliferation and DNA synthesis of PDLC, which may be accomplished by accelerating G 1 transition.
