摘要
目的 :探讨肾小球足细胞损伤在脂质肾损害发病中的可能作用。方法 :用含 4 %胆固醇和 1%胆酸钠的高脂饲料饲喂大鼠建立高脂模型 ,观察高脂组和正常组大鼠血脂、尿蛋白、肾功能及肾脏病理学改变 ,运用免疫组化技术检测肾小球足细胞损伤标志蛋白 -结蛋白表达 ,并进行相关分析。结果 :高脂组大鼠总胆固醇、低密度脂蛋白、血肌酐和尿蛋白均显著升高 (P <0 .0 1) ,肾小球系膜基质增生明显 (P <0 .0 1) ,肾小球内结蛋白表达上调 ,透射电镜下可观察到肾小球足细胞损伤表现。结蛋白表达与血脂、血肌酐、尿蛋白、肾小球系膜基质增生之间存在显著正相关性。结论
Objective: To investigate the possible effect of glomerular podocyte injury on lipid induced nephropathy in dietary induced hypercholesterolemic rats Methods: Dietary induced hypercholestero lemia was induced in male Wistar rats by giving them 4% cholesterol and 1% cholic acid diet for four months Serum lipid,urinary protein excretion, renal function and renal pathological changes were detected in the hypercholesterolemic and control rats Immunohistochemistry was used to detect the expression of desmin,a marker of podocyte injury At the same time,the correlation analysis were used Results: Serum total cholesterol and low density lipoprotein, serum creatinine,urinary protein excretion were increased significantly in hypercholesterolemic rats,accompanied with increased expansion of mesangial matrix and expression of desmin Podocyte ultrastructural changes were detected under electronic microscopy The increased expression of desmin showed a strong correlation with serum lipid, serum creatinine, urinary protein excretion and expansion of mesangial matrix Conclusion: Glomerular podocyte plays an important role in the pathogenesis of renal injury in lipid induced nephropathy
