摘要
目的 研究房水的免疫抑制功能,分析转化生长因子-β2(TGF-β2)在其中的作用。方法 分别用正常房水、酸化房水和加有TGF-β2抗体的酸化房水作用于淋巴细胞,通过 MTT比色法测定各种处理因素下的房水对脐带血淋巴细胞增殖的抑制作用,并计算抑制率。结果 正常房水可抑制淋巴细胞的增殖,抑制率为7. 87%;酸化房水的抑制作用明显加强,抑制率提高到 51. 53%;TGF-β2抗体可阻断酸化房水的活性,加入TGF-β2抗体后,抑制率下降到 9. 7%。结论 房水具有免疫抑制功能,可抑制淋巴细胞的增殖,而TGF-β2是产生这一作用的分子学基础。
Objective To analyse immunosuppressive properties in human aqueous humor. Methods Normal aqueous humor, acidification aqueous humor and acidification aqueous humor with TGF-β2 neutralizing antibody were cultured in lymphocytes stimulated by IL-1 in the presence of PHA, respectively. The proliferation rates of lymphocytes were measured with MTT staining colorimetry. Results It was found that aqueous humor could inhibit lymphocytes proliferation, the inhibition rates of normal aqueous humor, acidification aqueous humor and acidification aqueous humor with TGF-β2 neutralizing antibody were 7. 87% ,51. 53% and 9. 7% Respectively. Conclusion Aqueous humor has immunosuppressive effects and TGF-β2 presents in normal aqueous humor and this cytokine contributes to the immunosuppressive properties in aqueous humor.
