摘要
目的 观察吐温 80合并温热对人膀胱癌EJ细胞的PCNA和CyclinD1表达的影响。方法 应用免疫细胞化学和Western 印迹法等方法 ,检测EJ细胞经吐温 80合并 41℃温热作用后不同时间 (0h、16h)PCNA和CyclinD1表达的改变。结果 免疫细胞化学和Western 印迹法检测结果显示 ,PCNA和CyclinD1在对照组均为强阳性 ;加热组和加药组表达略有减少 ;而吐温 80合并温热作用后 ,人膀胱癌EJ细胞的PCNA和CyclinD1表达均受到显著的抑制。结论 吐温 80合并温热可显著降低人膀胱癌EJ细胞的增殖能力。
Objective To study the influences of Tween 80 combined with hyperthermia on the expressions of PCNA and Cyclin D 1 in human bladder cancer cell line EJ.Methods Tween 80 in combination with hyperthermia (41 ℃) was administered to EJ cells. The protein expression levels of PCNA and Cyclin D 1 were assayed by immuno cytochemistry and Western blotting method.Results Combined with Tween 80, hyperthermia(41 ℃) could dramatically decrease the expressions of PCNA and Cyclin D 1 ( P <0.01), with significant difference in comparison with that in hyperthmia or Tween 80 treatment alone.Conclusion The results demonstrated that Tween 80 combined with hyperthermia (41 ℃) can significantly inhibit the expressions of PCNA and Cyclin D 1 ,which might involve in the blocking of proliferation of EJ cells.
