摘要 目的 研究脑室注射Aβ1-4 0 对大鼠皮质、海马纤连蛋白基因表达的影响。方法 通过脑室注射Aβ1-4 0 ,建立大鼠AD模型 ,采用逆转录PCR方法研究纤连蛋白在皮质、海马的表达情况。结果 通过Morris氏水迷宫实验、脑片刚果红染色及胆碱乙酰转移酶表达水平等方面验证AD模型成功 ,在AD鼠皮质、海马纤连蛋白基因高表达。结论 纤连蛋白为整合素的配体 ,Aβ1-4 0 诱导其基因高表达可能介导Aβ1-4 0 与小胶质细胞的结合 ,激活小胶质细胞促进神经元死亡。 Objective To study the effect on gene expression of fibronectin (Fn) in cortex and hippocampus of rat after injection of Aβ 1 40 to cerebral ventricle.Methods The model of Alzheimer's disease of rat was established by injection of Aβ 1 40 to cerebral ventricle. RT?PCR technique was used to detect the differential expression of Fn in cortex and hippocampus.Results The model was verified successfully by Morris water maze, Congo red staining of brain slice, expression of gene cholinacetyl transferase (ChAT). The result of RT PACR for Fn mRNA revealed that it had higher expression in hippocampus and cortex of AD rats.Conclusion These findings suggest that Fn might play an important role in Aβ 1 40 binding to microglia and further activate it to induce the death of neurons.
