摘要
目的 研究丹参对兔严重烧伤早期 ,中性粒细胞 (PMN)与内皮细胞 (EC)粘附的作用及对血浆肿瘤坏死因子 -α(TNF -α)的影响。方法 2 4只日本大耳兔 ,建立兔 30 %Ⅲ度烧伤模型 ,随机分为丹参组和生理盐水组。分别于烧伤后即刻 ,2、4、8、16、2 4、4 8h ,取血分离获得PMN ,与体外培养的兔血管EC作用测定PMN -EC的粘附率 ,并测定各时点血浆TNF -α值。结果 烧伤后即刻生理盐水组与丹参组粘附率均升高 ,生理盐水组伤后 4h达高峰 ,丹参组 2、4、8、16、2 4、4 8h各时点的粘附率均低于生理盐水组 (P <0 0 5 )。生理盐水组于伤后 2hTNF -α开始升高 ,8h达高峰 ,4 8h仍高于正常 ,丹参组 2、4、8、16、2 4、4 8h各时点的TNF -α的值低于生理盐水组 (P <0 0 5 )。结论 丹参能降低严重烧伤后血浆TNF -α的水平 ,进而抑制PMN -EC粘附 。
Objective:To study the influence of Dan-shen Root to neutrophil-endothelial cell adhesiveness and the blood plasma level of tumor necrosis factor-α(TNF-α)of serious burn injury in earlier period. Methods: 30% Ⅲ drgree suffering burns rabbit model was made and randomly divided into Dan-shen group and saline group. Separately administered Dan-shen Root and saline and observed the changes of neutrophil-endothelial cell adhesiveness ratio and the contents of blood plasma of TNF-α instanly and in 2, 4, 8, 16, 24 and 48 hours after seriously burning. Result: After seriously burning, the neutrophil-endothelial cell adhesiveness ratios of two groups wese higher and in 2, 4, 8, 16, 24 and 48 hour after burning, the neutrophil-endothelial cell adhesiveness ratios and the contents of blood plasma of TNF-α of Dan-shen group were lower than that of saline group (P<0 05). Conclusion: Dan-shen Root through cutting down the blood plasma level of TNF-α and resisting neutrophil-endothelial cell adhesiveness of serious burn injury in earlier period to improve the microcirculation and lighten the pathologic damage.
