兔角膜植入改性HEMA人工角膜支架材料的实验研究 被引量:14

摘要 目的 :评价兔角膜层间植入改性聚羟乙基丙烯酸甲酯 [poly(2 hydroxyethylmethacrylate) ,PHEMA]孔隙性材料的生物学反应。方法 :选用新西兰白兔 ,将材料植入角膜板层口袋内 ,定期观察。结果 :植入后材料全部稳定存留于角膜内 ,现最长达 6个月以上。未见材料排出、白内障、视网膜脱离、角膜后膜等并发症。术后并发症包括睑缘红肿 ,畏光 ,流泪等刺激症状 ,均于术后 1周内消失 ;组织学提示材料孔隙内有多量的成纤维细胞并有胶原等细胞外基质沉积。电镜检查材料周围角膜细胞数量增多 ,排列规则 ,功能活跃 ;材料内见有纤维细胞 ,类似正常的组织愈合过程。结论 :结果提示此新型孔隙材料是人工角膜支架的理想材料 ;由于该材料还可以制作成光学性能良好的光学部 ,故可望制成一体式人工角膜。 Objective:to evaluate the biological response in vivo to poly 2-hydroxyethyl methacrylate(PHEMA) material for the use as the skirt of a keratoprosthesis. Methods:the porous discs of PHEMA were implanted into interlamellar pockets. The corneal tissues were evaluated clinically, histologically and ultrastructurally. Results: all of the implants have been retained, with no severe complications such as cataract, retrocorneal membrane formation, retinal detachment. Keratocytes migrationinto theimplants and depositionof complex extracellular matrix in the discs were found.The keratocytesaround the discs looked healthy and functionally activeby TEM. The migration of cells into the discs mimicked the scenario of wound repair. Conclusion:The moderated PHEMA is asuitable material for the skirt of one-piece keratoprosthesis.
