反胶束中枯草杆菌中性蛋白酶的活力研究 被引量:4

摘要 系统地研究了枯草杆菌AS1 398中性蛋白酶在AOT/异辛烷反胶束体系中的催化行为 ,研究底物浓度、表面活性剂浓度、水含量、pH值、温度、盐的浓度及种类对酶活力的影响 .结果表明 :酶在反胶束体系中表现出“超活性” ,且作用于胶束膜 .在AOT/异辛烷浓度为 0 0 8g/mL时 ,影响酶活力大小主次因素为 :pH值 >温度 >盐浓度W0 .最佳酶活力条件 :5 0℃ ,0 2mol/LKCl液 ,pH值 8 0 ,W0 值 1 1 6,此时酶活力为 6 2 6× 1 0 5u/g . The catalysis of neutrase AS1.398 incorporated in AOT/iso-octane was investigated. The effects of the substrate concentrations, waterpool( W 0 ),pH,temperature, kinds of salt and concentrations were studied in detail.It indicated that enzyme showed 'superactivity' and acted on the membrane of reverse micelles. When the concentration of AOT/iso-octane was 0.08 g/mL,factors affecting the activity of the enzyme were in sequence as:pH value>temperature>salt concentration> W 0 .The maximal activity was 6.26×10 5 u/g at 50℃,0 2 mol/L KCl,pH8.0 and W 0 value 11.6
