抗人精子单克隆抗体对生殖的影响

摘要 本文报告9株抗人精子单克隆抗体在体外对生殖的影响。TAT、SIT、SCMC和精子—地鼠去透明带穿卵实验表明:3株IgM类抗人精子单克隆抗体主要引起精子的头—头凝集,对精子有较强的补体依赖性细胞毒作用;6株IgG类抗人精子单克隆抗体主要引起精子的尾—尾凝集,并能阻止精子在宫颈粘液中的前向性运动;4/9株抗人精子单克隆抗体对精子穿卵有明显抑制作用,且两株联合应用有协同效应,提示有效而可靠的精子避孕疫苗应具有多个抗原决定簇。 The influence of nine strains of monoelonal antibodies against human sperm on repro- duction were evaluated in vitro. The results of TAT, SIT, SCMC and sperm-hamster egg pcnetration assay show that: (1)three strain of IgM produce mainly H-H agglutination of spermatozoa and its complement-dependent cytotoxicity to sperm is stronger. (2)six strains of IgG mainly produce T-T agglutination of spermatozoa and being able to prevent the spermatozoa from forward movement in the cervical mucus. (3) four strains of monoclonal antibodies against human spermatozoa inhibited the sperm-hamster egg penetration significantly and there is synergistic effect while two strains being combined. It is suggested that the effective and relible sperm contraceptive vaccine have multiple antigenic determinants.
