摘要 目的 提取牛骨形态发生蛋白 ,并观察其对人骨髓基质细胞的体外成骨分化作用 ,为将骨形态发生蛋白与骨髓基质细胞用于进一步的骨缺损修复研究提供实验依据。方法 从小牛骨中提取骨形态发生蛋白 ,配制条件培养液 ,并作用于培养状态的人骨髓基质细胞 ,染色检测细胞碱性磷酸酶、Ⅰ型胶原表达情况 ,放免法测定培养液中骨钙素含量。结果 人骨髓基质细胞经诱导培养后 ,碱性磷酸酶、Ⅰ型胶原、骨钙素表达明显增加。结论 体外培养时 ,天然骨形态发生蛋白可促进人骨髓基质细胞的成骨分化。 Objective To investigate the effects of bone morphogenetic proteins (BMP S) on the differentiation of human marrow stromal cells (MSC S) into osteoblasts in vitro for experimental basis of MSCs in bone defect reparation. Methods BMP S were extracted from one-year-old bovine bone and then dissolved into RPMI1640 medium. Human MSC S were cultured with the prepared special medium, and then stained for alkaline phosphatase (ALP) and typeⅠ collagen, measured for the amount of osteocalcin in the culture medium. Results After human MSC S were cultured with the prepared medium, the expressions of ALP and typeⅠ collagen and the amount of osteocalcin were increased greatly. Conclusion Natural BMP S can promote human MSC S differentiated into osteoblasts in vitro .
