组织工程化关节软骨细胞凋亡的研究 被引量:9

摘要 目的 观察离心管构建的组织工程软骨细胞凋亡及细胞增殖情况。方法 对离心管构建 3周的组织工程软骨进行透射电镜、流式细胞仪检测和3 H TdR掺入测定 ,与 3周龄兔关节软骨对照。结果 透射电镜检查组织工程软骨有 5 %左右的细胞凋亡 ,正常关节软骨未见明显的细胞凋亡 ;流式细胞仪显示软骨细胞有 8.5 %左右的亚二倍体细胞 ,正常关节软骨有 2 .4%左右的亚二倍体细胞。3 H TdR掺入测定显示细胞增殖能力较正常关节软骨明显增强。结论 培养 3周的组织工程软骨细胞凋亡与细胞增殖均较正常软骨增强。 Objective To observe the apoptosis and proliferation of chondrocytes in tissue engineered articular cartilage 3 weeks after construction with centrifuge tube. Methods Tissue engineered articular cartilage constructed by centrifuge tube were studied with electron microcopy, TUNEL, 3H-TdR incorporation and flow cytometry. Results Nearly 5% apoptotic cells were found in the engineered articular cartilage under a microscope which were similar with that obtained by flow cytometry. The chondrocytes in tissue engineered articular cartilage had more 3H-TdR incorporation than the articular chondrocytes from 3-week-old rabbits. Conclusion The chondrocytes in 3-week tissue engineered articular cartilage have stronger potency in both proliferation and apoptosis than the cells in normal articular cartilage.
