摘要
目的 探讨大鼠坐骨神经损伤后再生过程结构形态三维重建的方法 ,三维重现再生神经通过损伤界面和吻合口的过程及规律。方法 用硅胶管桥接 6 0只大鼠左侧坐骨神经长 6mm缺损 ,术后 3、7、15、30、6 0、90d取坐骨神经远近端各 1cm ,石蜡包埋 ,连续切片 ,HE、硝酸银 luxol快蓝染色 ,显微摄像输入微机后进行图像配准、二维综合处理 ,最终在SGI三维图形工作站上完成三维重建和显示。结果 三维图像显示新生神经纤维与正常神经纤维相比有郎飞氏结尚未形成、排列紊乱、髓鞘较薄、轴索较细、神经膜管形成等差异 ,胶原纤维大量增生阻碍了新生神经纤维的生长。结论 利用计算机图像重建技术 ,采用三维图像的形式对外周神经再生过程和再生规律进行可视化研究 ,是一种有效的方法。
Objective To explore the method for 3-D reconstruction of morphological structure of regenerated sciatic nerve of rats after injury in order to tridimensionally depict the process and rule of damaged nerve regenerating across the lesion interface and anastomosing. Methods A total of 60 Wistar rats were divided randomly into 6 groups. The sciatic nerve was transected by 6 mm and then repaired with silicone tube. The sciatic nerve pieces of 1cm long in the close and remote ends were cut 3, 7, 15, 30, 60, and 90 d respectively after injury, then embedded with paraffin, serially sliced, dyed with HE and silver-nitrate fast blue. The images of slices were taken by micro-photograph system and inputted into computer by which the image matching and 2-D integrate processing was completed. The 3-D reconstruction and display was accomplished in SGI workstation. Results Compared with normal nerve fibers, some difference in the neonatal nerve fiber was showed by the 3-D images. Ranvier nodes were not formed and in turbulence. Myelin sheath and axis-cylinder were much thin. No neurolemmal tube was found. Conclusion It is a practical method to study the visualization of peripheral nerve regeneration with 3-D images by means of computer 3-D reconstruction technique.
