摘要
本研究通过生物信息学和分子生物学相结合的方法成功克隆了一个新的锌指蛋白基因ZNF333cDNA。ZNF333基因包含 14个外显子 ,编码 6 6 5个氨基酸。蛋白质N端包含 2个保守的KRAB A盒 ,2个多态的KRAB B盒 ,而C端包含 10个重复的C2 H2 型锌指基序。ZNF333的表达在心脏组织中最高 ,在其他组织中也可检测到表达。同时还发现该基因存在选择性剪切形式。通过与人类基因组框架图的比较将ZNF333定位在 19p13 1。
A novel human KRAB zinc finger protein gene, ZNF333 , was identified with computerized and experimental methods. The putative ZNF333 protein encodes 665 amino acids and contains two conserved KRAB A boxes, two divergent KRAB B boxes at the N terminus and 10 C 2H 2 zinc finger motifs at the C terminus. An alternatively spliced isoform of ZNF333 was also characterized. ZNF333 expression is the most abundant in heart but detectable in other tissues as well, including human fetal limb. According to the human genome working draft sequences, ZNF333 gene is localized at 19p13.1.
