摘要
探讨血红素氧合酶 - 1(HO- 1)在豚鼠哮喘模型中的表达特点。将 2 4只豚鼠随机分为 4组 ,每组 6只 :1正常对照组 (A组 ) ;2哮喘即刻激发组 (B组 ) ;3哮喘发作 1周组 (C组 ) ;4哮喘发作 2周组 (D组 )。测定全血一氧化碳血红蛋白 (COHb)的百分比含量并观察气道壁嗜酸性粒细胞 (EOS)浸润情况 ;用免疫组织化学染色方法观察 HO- 1在豚鼠哮喘模型肺组织中的表达特点。发现 HO- 1主要表达在气道上皮细胞 ,各组 HO- 1阳性表达的平均吸光度分别为 0 .0 32±0 .0 0 4、 0 .112± 0 .0 11、 0 .12 3± 0 .0 11和 0 .135± 0 .0 12。哮喘各组 (B、C、D组 ) HO- 1的表达水平均显著高于 A组 (P<0 .0 1)。HO- 1的表达水平与全血 COHb的百分比含量及气道壁 EOS计数均呈显著正相关 (分别为 r=0 .88,P<0 .0 0 1和 r=0 .86 ,P<0 .0 0 1)。结果表明哮喘豚鼠气道上皮细胞 HO- 1的表达水平显著增加 ,提示 HO-
The expression of heme oxygenase 1 (HO 1) in asthmatic guinea pigs was investigated. Twenty four guinea pigs were randomly divided into 4 groups of 6 each: normal control group (group A), asthmatic instant stimulationer group (group B), one week asthmatic group (group C) and two week asthmatic group (group D). The pacent content of blood carbon monoxide Hb (COHb) and eosinophilic inflammation of airway wall were observed. The expression of HO 1 in lung tissue was detected by using immunohistochemical staining. The results showed that the expression of HO 1 was mainly located in airway epithelium in the 4 groups (A, B, C and D) with the optical densities being 0 032±0 004, 0 112±0 011, 0 123±0 011 and 0 135±0 012, respectively. The asthmatic groups (groups B, C, D) all showed higher optical densities than that of the normal control group ( P <0 01). There was a significant positive correlation between the optical densities of HO 1 in epithelium and the percent content of blood COHb ( r =0 88, P <0 001) or the amount of eosinophil in airway wall ( r =0 86, P <0 001). These results showed that the expression of HO 1 was increased significantly in the airways of the asthmatic guinea pigs, suggesting HO 1 might play a significant role in the pathogenesis of asthma.
