摘要
目的 研究丹参对缺氧复氧后心室肌细胞L 型钙通道电流 (L Ica)的影响。方法 应用膜片钳全细胞记录技术。结果 经过 2 0min的缺氧以及 5min的复氧后 ,5 0、10 0、2 0 0mg/L丹参分别将L Ica的峰值从 (6 6 7 9± 15 8 7)pA减小至 (5 37 1± 12 1 3) pA、(4 97 2± 47 9) pA (P <0 0 5 ,n =5 )、(34 3 8± 73 1) pA(P <0 0 1,n =5 )。 结论 丹参能有效地减少缺氧复氧后心室肌细胞的L Ica ,这种作用可能是其抗心肌缺血以及缺血再灌性心律失常的机制之一。
Objective To investigate the effect of Danshen on L-type calcium channel currents (L-Ica) in rabbit ventricular myocytes exposed to hypoxia. Methods Whole cell patch clamp technique. Results After 20 min hypoxia and 5 min reoxygenation, 50?100?200 mg/L Danshen reduced the L-Ica from (667.9±158.7) pA to (537.1±121.3) pA,(497.2±47.9) pA (P<0.05,n=5)?(343.8±73.1) pA (P<0.01,n=5) respectively. Conclusion Danshen can significantly reduce the L-Ica during early reoxygenation, which may contribute to its effect on anti-myocardial ischemia and anti-reperfusion arrhythmia.
