摘要
目的 :观察从柴胡中提取的单体成分柴胡皂甙d诱导白血病细胞株K562细胞凋亡及对其基因表达的影响。方法 :分别用 5μg/ml、7.5μg/ml、1 0 μg/ml浓度的SSd作用于K562细胞株 ,于药物作用后不同时间段 (1 2 ,2 4 ,36 ,48,60h) ,荧光染色法观察基凋亡状况、间接免疫荧光法测Bcl- 2表达情况。结果 :荧光染色结果显示 :K562在 1 0 μg/ml处理 2 4h后 ,开始出现凋亡小体 ,48h后出现大量死细胞。间接免疫荧光法测Bcl- 2表达 ;经SSd作用后K562细胞Bcl-
Objective:To research the effect of SSd on the cell apoptosis and gene expression of K562 cells. Method:Apoptosis of K562 cells was analyzed by fluorescent stain technique and the expression of Bcl-2 was determined by immunoflourescent antibody technique after the treatment of different density of SSd (5μg/ml?7.5μg/ml?10μg/ml) at different time (12,24,36,48,60h).Results:The fluorescent stain showed that after 24 hours' treatment with 10μg/ml SSd,apoptosis bodies appeared in the K562 cells.After 48 hours,a great deal of dead cells appeared. The expression of Bcl-2 was decreased by ssd.Conclusion:SSd could down-regulate the expression of Bcl-2 and induce apoptosis of K562 cell lines.
