压力超负荷时bcl-2蛋白表达对兔心肌细胞凋亡的影响

摘要 目的 :探讨压力超负荷时兔左室心肌细胞bcl- 2蛋白表达对兔左室心肌细胞凋亡的影响。方法 :采取末端脱氧核苷酸转移酶介导的DUTP -生物素平移末端标记 (TUNEL)技术和链酶亲和素 -生物素 -过氧化物酶复合物 (SABC)免疫组化方法 ,观察持续压力超负荷下兔左室心肌细胞凋亡的动态变化和bcl- 2蛋白的表达。结果 :对照组罕见凋亡细胞 ,表达bcl- 2蛋白的心肌细胞少而散在。术后凋亡阳性心肌细胞数和bcl- 2蛋白阳性表达心肌细胞数迅速增加 ,两者在心衰组最高。凋亡阳性心肌细胞数在术后第 3天与术后第 7天之间存在一个峰值。凋亡阳性心肌细胞和bcl- 2蛋白阳性心肌细胞主要分布在心肌深层、乳头肌和中层心肌。结论 :慢性压力超负荷持续存在时 ,bcl- 2蛋白阳性表达心肌细胞数存在动态变化 ,其分布与凋亡心肌细胞分布相似 。 Objective:To study the effect of bcl-2 protein expression on myocyte apoptosis during chronic pressure overload in left ventricle of rabbit.Methods:We investigated the dynamic changes of myocyte apoptosis and the expression of bcl-2 protein in left ventricle of chronic pressure overload-induced rabbit,using in situ terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) and strept avidin-biotin-enzyme complex (SABC) immunohistochemistry.Results:There was a little apoptosis and expression of bcl-2 protein on myocytes in control group.The apoptosis and bcl-2 protein expression of myocyte in left ventricle were increased after operation.The number of day 3 group was higher than the number of day 7 group and day 1 group.The number of apoptosis and expression bcl-2 protein of myocytes were the highest.The apoptosis and bcl-2 protein expression of myocytes were the highest.The myocytes of apoptosis and expression of bcl-2 protein were found in suberdocardial layer,papillary muscle and mesocardial layer.Conclusions:During chronic pressure overload,the expression of bcl-2 protein in myocyte had dynamic changes,its distribution was simliar to the distribution of myocyte apoptosis,and bcl-2 protein was the major antiapoptotic protein.
