摘要 目的 对慢性接触镉大鼠投与GSH、维生素C和DMPS ,探讨这 3种物质对镉慢性毒性的影响。方法 连续 1 1周给 4个实验组大鼠皮下注射CdCl2 5μmol/kg,对照组大鼠皮下注射生理盐水。然后给实验组大鼠分别腹腔注射GSH、维生素C、DMPS和生理盐水共 2周。对照组大鼠在相同时间腹腔注射生理盐水。最后一次注射后 2 4h ,收集大鼠 2 4h尿液 ,处死大鼠 ,采集血液、肝脏和肾皮质样品。测定肝脏、肾皮质的GSH、MDA、蛋白质和镉含量 ,GSH Px活性以及尿镉含量 ,测定血MDA含量、GSH Px活性和血红蛋白含量。结果 单纯染镉组大鼠肝脏镉浓度明显高于DMPS投与组 ,肾镉和尿镉含量明显低于DMPS投与组 ;各实验组大鼠肝脏和肾皮质GSH含量均明显高于对照组 ,单纯染镉组GSH含量明显高于GSH、维生素C和DMPS投与组 ;单纯染镉组大鼠肝脏、肾皮质和血GSH Px活性明显低于其他各组 ;GSH、维生素C和DMPS投与组大鼠肝脏、肾皮质、血MDA含量与单纯染镉组比均有明显降低。结论 GSH、维生素C和DMPS对镉慢性毒性损害的恢复有一定的促进作用 ,GSH和DMPS还可能促进镉从尿中排泄。 Objective\ To study effects of DMPS,GSH and Vit\|C on chronic cadmium toxicity in rats.Methods\ Fifty rats were randomly divided into five groups.Four groups were given subcutaneous injections of 5 μmol/kg CdCl 2 and the rest group with 0\^9% NaCl for 11 weeks.Beginning from the 12 th week,the 2 nd group was intraperitoneally injected with 0\^9% NaCl,the 3 rd group with 1 mmol/kg GSH,the 4 th group with 4 mmol/kg Vit\|C,and the 5 th group with 0\^2 mmol/kg DMPS for 2 weeks.24 hours after the last injection,urinary samples were collected.And 48 hours after the last injection,rats were sacrificed,and the liver,kidney cortex and blood samples were collected.Cd concentration in liver,kidney and urine were determined.The GSH\|Px activity,protein contents,GSH and MDA concentrations in liver and renal cortex were determined,and GSH\|Px activity,protein contents,MDA concentrations in blood were also determined.Results\ Cd concentration in liver of each group was lower than that of the group administrated with Cd alone,while the results were reversed in kidney.DMPS,GSH and Vit\|C administration significantly increased GSH contents both in liver and kidney,The GSH concentration in the group given Cd alone was higher than those of other three experimental groups.The GSH\|Px activities in DMPS,GSH and Vit\|C groups were not different from that of the control group,However,the GSH\|Px activity in the group given Cd alone was lower than those of other three experimental groups.After administration of GSH,Vit\|C and DMPS,the contents of MDA in liver,kidney and blood samples decreased significantly.Conclusions\ Cd administration resulted in liver and kidney damages in SD rats.DMPS,GSH and Vit\|C might have protective effects on Cd induced hepatic and renal toxicity at different levels.
