苯并[a]芘对小鼠神经毒性的光镜、电镜及行为学研究 被引量:12

摘要 目的 研究苯并 [a]芘对小鼠神经系统的损伤情况。方法 将 50只昆明小鼠分为 5组 ,每组 1 0只 ,染毒组用苯并 [a]芘的植物油溶剂进行腹腔注射处理 ,每次剂量分别为 7 8,3 2 ,1 3mg/kg,每周 4次 ;溶剂对照组用植物油溶剂作平行处理 ;空白对照组不施加处理因素。实验中观察一般情况 ,于实验第2 1、 35、 49、 63天各进行 1次行为学实验 ,实验 1 0周后取小鼠脑组织、脊髓与坐骨神经制作光镜及电镜切片并观察。结果 发现苯并 [a]芘染毒组光镜及电镜下可见明显的神经组织损伤 ,行为学测试中游泳实验记分较对照组低 ,而溶剂对照组及空白对照组组织结构未见异常 ,行为学实验正常。结论 苯并 [a]芘具有神经毒性。 Objective\ To study the neurotoxicity of benzopyrene (BP).Methods\ 50 Kunming mice were equally divided into 5 groups.The experiment groups were intraperitoneally injected with BP dissolved in vegetable oil in dosages of 7\^8,3\^2 and 1\^3 mg/kg per injection,4 times a week.The vehicle control group was dosed with vegetable oil,and the standard control group without any additional treatment.In the experiment,the general toxicity reaction and behavioral reaction were observed.On the 21\+\{st\},35\+\{th\},49\+\{th\} and 63 rd day,the behavioral damages of mice were examined.10 weeks after the experiment,the brain,spinal cord and osiscii nerves were removed.Pathological damages of optical slices and electron\|microscope slices from brain(cortex,hippocampi,cerebellum and brainstem),spinal cord and osiscii nerves of all 5 groups were observed.Results\ From the behavioral experiment,the dosed groups showed lower swimming scores than those of the control groups.In the dosed groups,damages could be seen in both optical slices and electron\|microscope slices,however,in the standard control group and vehicle control group,no significant damage could be seen.Conclusions\ BP could damage the nerve system of mice.
