摘要 目的 研究较长期染锰对大鼠脑、血有核细胞的线粒体功能的影响 ,以探讨脑、血有核细胞线粒体功能异常在锰神经毒性中的作用。方法 健康Wistar大鼠分为高剂量 (2 5mg/kg)、中剂量(1 5mg/kg)、低剂量 (5mg/kg)及对照组 ,腹腔注射MnCl2 ·4H2 O 3个月后处死 ,分别检测其脑、血有核细胞线粒体的呼吸控制比 (RCR)、ATP生成、膜电位差的变化。结果 与对照组相比 ,高、中剂量组大鼠脑线粒体功能的 3个指标 (RCR、ATP、膜电位差 )均显著降低 ,大鼠血有核细胞线粒体的呼吸控制比、ATP生成显著降低 ,高剂量组大鼠血有核细胞线粒体的膜电位差显著降低。此外 ,染毒结束时中、高剂量组大鼠平均体重明显低于对照组 ,脑 /体比值明显高于对照组。其余各组脑、血有核细胞线粒体功能改变差异无显著性。结论 亚慢性染锰对大鼠脑。 Objective\ To study the effect of long\|term Mn exposure on mitochondrial function of rat brain and hemakaryocyte,and to explore the action of mitochondrial dysfunction of brain and hemakaryocyte.Methods\ Wistar rats were divided into four groups:high,medium,and low dose groups and the control group,Rats were received ip.injection of MnCl 2\54H 2O once a day for three months.The brain and hemakaryocyte mitochondrial functions including RCR,ATP,and membrane potential were examined after sacrifice.Results\ As compared with the control group,both RCR,ATP and membrane potential of rat brain,and RCR,ATP of rat hemakaryocyte mitochondrial in high and medium groups showed marked reduction.In high dose group,the membrane potential of rat hemakaryocyte mitochondrial also showed marked reduction.In addition,three months later,the rat weight in medium and high dose groups was significantly decreased and the ratio of brain/body weight was significantly increased.The rest did not show statistically significant difference.Conclusions\ Sub\|acute Mn exposure might cause mitochondrial dysfunction of rat brain and hemakaryocyte.
