脐带血清代替GM-CSF对树突状细胞分化和功能的影响

摘要 目的 :观察脐带血清 (Umbilicalcordserum ,UCS)取代粒单细胞集落刺激因子 (GM -CSF) ,对体外培养体系中树突状细胞分化和功能的影响。方法 :用脐带血清代替GM -CSF培养出树突状细胞 ,通过流式细胞仪检测细胞表型 ,MTT法检测混合淋巴细胞反应的能力。结果 :脐带血清代替GM -CSF培养的DC(UCS -DC)表面CD83、CD86和HLA -DR的表达率比常规组DC明显升高 ;而CD1a表达率降低 ,对混合淋巴细胞反应的刺激能力UCS -DC与常规组均较对照组明显增强 ,但二者之间比较未达到统计学意义。结论 :含丰富细胞因子的脐带血清能代替GM -CSF产生出表型更成熟、有较强刺激同种异体T淋巴细胞增殖能力的DC。 Objective:To observe the pherotype and function of DC when GM-CSF was replaced by umbilical cord serum in the DC medium. Methods:Dendritic cells were cultured with umbilical cord serum instead of GM-CSF, the phenotype of DC was detected by FACS and the antigen presenting ability of DC was measured in allo-MLR by MTT assay.Results:Dendritic cells grown in the group in which GM-CSF was replaced by umbilical cord serum (UCS-DC) had higher expression rate of CD86, CD83 and HLA-DR than FCS-DC(P<0.05), and their expression of CD1a was lower than that of FCS-DC. Their ability to induce T cell proliferation had no difference. Conclusion:The umbilical cord serum rich in cytokines, in some extent, it could replace GM-CSF in the course of dendritic cells-culturing, and such cultured dendritic cells had more mature phenotype, and higher ability of stimulating T cells proliferation.
