神经生长因子及其受体家族同步下调在胃癌发生中的作用 被引量:6

Effects of nerve growth factor and Trk families down regulation on gastric cancer

摘要 目的研究神经生长因子(NGF)及其受体家族在胃癌组织及其相对应正常胃黏膜中mRNA的表达,探讨它们在胃癌发生中的作用。方法5例胃癌患者癌组织mRNA及相对应的正常胃黏膜组织mRNA,等量混合分别构成胃癌组织mRNA组及正常胃黏膜mRNA组,α-33PdATP同位素标记反转录cDNA作探针,采用Microarray基因微阵列杂交技术,检测胃癌与相对应正常胃黏膜间NGF及其受体Trk家族mRNA表达,分析其表达差异。结果NGF及其受体Trk家族在胃癌组织及其相对应正常胃黏膜中均有不同程度的表达。与相对应正常胃黏膜相比,胃癌组织中NGF、BDNF、NT-3、NT-4/5、NT-6及受体TrkA、B、C均同时下调,NGF家族下调程度大于Trk家族,其中NT-3及BDNF下调程度最大。Trk家族中TrkC下调程度最大。结论与NGF及Trk家族在其它肿瘤组织中表达上调不同,NGF家族及Trk家族在胃癌组织表达下调,它们在胃癌发生中极可能是通过另一新的Ras或Raf信号传导途经,而发挥独特的促凋亡作用。 Objective To examine mRNA expression of nerve growth factor(NGF) and Trk families in gastric cancer and counterpart normal gastric mucosa, and investigate effects of NGF and Trk families on the tumorigenesis of gastric cancer. Methods Counterpart normal gastric mucosa mRNA mixture and gastric cancer tissues mRNA mixture from five patients were reversely transcripted into cDNA with dNTP in isotope labeling respectively. mRNA expression of NGF and Trk families were detected in both gastric cancer and counterpart normal gastric mucosa using cDNA microarray. mRNA expression difference between gastric cancer and counterpart normal gastric mucosa was analyzed. Results NGF and Trk families mRNA expressed in different degrees. NGF,BDNF,NT 3,NT 4/5,NT 6 and TrkA,TrkB,TrkC were down regulated simultaneously in gastric cancer with comparison to counterpart normal gastric mucosa. Down regulation in NGF families presented greater than that in Trk families. In gastric cancer, NT 3 and BDNF decreased more than 90% and TrkC down regulated more than 75% as compared to counterpart normal gastric mucosa. Conclusions Other than up regulation of NGF and Trk families in other tumors, NGF and Trk families are down regulated in gastric cancer. NGF and Trk families may play an unique apoptosis role in gastric cancer by another new Ras or Raf signal transducted pathway.
