摘要
采用反相HPLC法测定白花前胡根中醚提取物的有效成分 [3′(S) 当归酰氧基 ,4′(S) 乙酰氧基 3′ ,4′ 双氢邪蒿内酯 ,Pd Ia]的含量。峰面积与Pd Ia含量在 0 .0 0 15 0 5~ 0 .15 0 5mg·ml-1范围内呈线性关系。回归方程为C(mg·ml-1) =5 .8× 10 -8A - 5 .0 2 5× 10 -4 。前胡样品中Pd Ia的含量为 0 .0 77%。
Reversed phase HPLC determination of the effective component, 3′(S)-angeloyloxy-4′(S)-acetoxy-3′,4′-dihydroseslin (abbreviated as Pd-Ia) in roots of Baihua Qianhu was described. The Pd-Ia in the sample was first extracted with ether, and then further separated on a spherisorb 18 column (3.9mm×150mm) using CH 3OH-H 2O (80∶20) as the mobile phase and finally measured at 322nm with UV-detector. There was a good linear relationship between peak area A and concentration C of Pd-Ia in the range of 0.001505~0.1505mg·ml -1. The linear regression equation was found as follows: C=5.8×10 -8A-5.025×10 -4 where C=mg of Pd-Ia per ml of solution, A=peak area measured. And the value of corrlation coefficient was found to be 0.9998. The average content of Pd-Ia found by analyzing a typical sample of Baihua Qianhu was 0.077% (n=5).
